GLYCAN ARRAY SCREENING

Processing of Requests for Glycan Array Analysis by Core H
Core H has developed a process for receiving samples and reporting data that encompasses communication, sample handling, glycan array manufacture, standardized assays, data processing, data upload, and data dissemination. The Core H policy is to send all data from completed projects to the web site in a timely manner. Once the data is uploaded and 'finalized' it becomes available to members of the Consortium and 6 weeks later it is automatically made public on the Consortium Website. The current mechanism for handling requests is a multi-step process:
- Core H receives notification from Core A that a resource request has been submitted for Core H.
- Core H Director determines if Core H has the resources to support the request and notifies the Steering Committee via a comment on the website.
- Core H Director is notified after the SC meeting that the request is approved or conditionally approved.
- Core H director sends a Sample information Entry Form (SIEF) to the PI
- The completed SEIF contains enough information to define the Glycan (or Carbohydrate) Binding Protein (GBP or cbp) for entry into the database.
- The complete SIEF also contains sufficient information to permit the Core H Director and staff to design the analysis.
- When the completed SIEF is received:
- Core H Director Enters the cbp information and the cbp ID number onto the website and stars a folder under the communicating investigator's name and the Request number. All requests are followed by the request number that was assigned when the PI initiated the request. It is very important for the PI to include that request number on the subject line of all communications (especially e-mail and shipping information) with Core H so that we can keep track of our communication.
- Core H Director and staff complete a Core H Glycan Array Processing form that describes the experiment for each cbp being analyzed. The Glycan Array Processing Form becomes a written record of the analysis and is stored as a hard copy in a folder for future reference to support any publication of the data. The completed Glycan Array Processing Form identifies the request number, communicating investigator, sample names, exact experimental conditions used, and places for the Technical Staff to record all information regarding sample description, date and conditions of sample receipt and any deviations from the proposed protocol in the analysis.
- Core H Director then provides the Technical Staff with a folder containing the completed SIEF and the Glycan Array Processing Form.
- Shipment of sample(s) is scheduled only after receiving the completed SIEF form a request that has been approved by the Steering Committee.
- Sample is received
- The Technical Staff organizes the anticipated workload and schedules the assays.
- Each Friday and Monday, the Technical Staff and Core Director meet to discuss the analyses to make sure they agree on the conditions of the assays. The Core Director and Technical Staff are also in constant communication regarding status of samples and questions regarding deviations.
- When the assays are complete, the file with hard copies of the histogram and associated data are returned with SIEF and the Glycan Array Processing Form to the Core Director for Review and the results are sent by e-mail to the Participating Investigator.
- When the Core Director and Investigator agree that the data are interpretable, the data are uploaded to the website by the Technical Staff or the Core Director and made accessible to the PI if he/she is a member, Core H Coordinator and the Directors of other Cores. After one week (approximately), if there have been no issues raised, the data are finalized. Core H and the bioinformatics Core B are working on processes whereby results that are considered negative or uninterpretable will be uploaded in a non interactive format on the website so that it will be possible to know what cbps have been analyzed and the conditions that gave no interpretable results on the glycan array.
