Experiment to be conducted
:
The goal of this experiment is to determine the glycan receptor binding specificities of emerging influenza virus hemagglutinin (HA) proteins including avian H5N1 and swine-like H1N1. His-tagged HA protein ectodomain has been produced using a baculovirus expression system and has been purified to >99% purity and is available immediately. In collaboration with the Consortium, we proposed to ship the protein immediately for analysis of its glycan binding profile. We believe there is an urgent need to perform this work for the HA protein from the emerging swine-origin H1N1 influenza that has reached pandemic status. A total of 2 HA proteins are available now (1 H1N1 and 1 H5N1), 1 HA protein should be available next week, and we may generate a 4th HA protein during the next month.

GBP being Addressed
:
The HA proteins contain a C-terminal His tag which can be used for binding of the primary antibody (a His-tag antibody).
Specifc aims being addressed
:
Define the specificity and affinity for carbohydrate ligands. Identify the ligand(s) that mediate GBP binding.
