Experiment to be conducted : Soluble lectins from Pseudomonas aeruginosa have been demonstrated to be involved in the adhesion to epithelial cells and in the infection process. PA-IL (LecA - galactose specific) and PA-IIL (LecB - fucose specific) have been extensively studied for their structure and inhibition and PA-IIL has been selected among the paradigms for GBP in the consortium. Nevertheless, their specificity was not tested on the recent version of the glycan array (specially PA-IL that was only tested on the first version of the plate). In collaboration with David Smith we undergo a complete specificity caracterisation of Pseudomonas aeruginosa soluble lectins including comparison with the ones from related opportunistic bacteria Chromobacterium violaceum,Burkholderia cenocepacia and Ralstonia solanacearum.

If yes, indicate the person responsible for inputing data into core B : David Smith
GBP being Addressed : All GBP are produced in recombinant form and labelled with Alexa fluor 488
Specifc aims being addressed : Define the specificity and affinity for carbohydrate ligands. Identify the ligand(s) that mediate GBP binding. Determine the structures of selected GBPs.
