Experiment to be conducted
:
We would like to see if we could do glycan array with rotavirus VP8. We have recently determined the X-ray structure of human rotavirus VP8. Rotavirus is the major pathogen of infantile gastroenteritis, which account for more than half million children deaths annually worldwide. VP8 is the domain of the rotavirus spike protein which has been implicated in hemagglutination and glycan binding. Human rotaviruses are not neuraminidase sensitive, and they are thought to bind to glycan with internal sialic acid such as GM1. We wanted to carry out glycan array to determine which glycan this human VP8 binds to and carry out crystallographic analysis of VP8 with that glycan. We already see interesting structural differences between our VP8 and other VP8 structures that have been published.
We have GST-tagged VP8 in large quantities for this experiment. We could ship the protein as early as next week.
