Experiment to be conducted : Previous studies have demonstrated an unprecedented utility of using the glycan microarray to elucidate galectin family member carbohydrate recognition properties. This has been most readily accomplished using a series of concentrations to determine the relative affinity of galectins for different glycans. This is of utmost importance considering that subtle differences in galectin recognition and glycan modification can significantly affect galectin signaling and cellular sensitivity. Although many studies have employed human galectin-1 in murine systems, very few studies have directly examined the biological activity of murine galectin-1. Our preliminary studies suggest that human galectin-1 and murine galectin-1 have distinct biological activities toward cells from different species. To explore this in more detail, we propose to examine human and mouse galectin-1 over a broad range of concentrations, at 10, 5, 1, 0.5 and 0.1 µM. As recent studies strongly suggest that galectins appear to display two physiologically relevant dynamic concentrations ranges, where 10-1 µM typically signals leukocytes, and 1 – 0.1 appears to regulate other biological functions, examining these particular concentrations should position us to observe important differences in each dynamic range, which can then be used to make predictions in unique biological systems, as done previously.
